A solution of 2-(4-chloro-2-methoxypyridin-3-yl)-6-(2-(dimethylamino)ethyl)-4-methylimidazo[4,5-f]isoindole-5,7(1H,6H)-dione in a mixture of 1,4-dioxane (14 mL) and concentrated HCl (2.5 mL) was heated at 45-50° C. for 14 h. After the end of reaction was established by TLC, the reaction mixture was concentrated under vacuum to afford a crude product as a yellow solid, which was used for the next step without further purification. Mass (ESI, positive) m/z 400.1 [M+H]+.
