Procedure

To a solution of 3-(5,6,7,8-tetrahydro-[1,8]naphthyridin-3-yl)-isonicotinic acid methyl ester (131 mg, 0.49 mmol) in DCM (30 mL) is added benzoyl isocyanate (107 mg, 0.73 mmol). The mixture is heated to reflux for 3 hrs. The solvent is then removed and the residue is dissolved in anhydrous MeOH (10 mL). Then K2CO3 (180 mg, 1.3 mmol) is added and the mixture is heated to reflux for 0.5 h. After cooling down, the organic solvent is evaporated. The residue is dissolved in water and washed with DCM. The water layer is separated and purified by neutral preparative HPLC to give 80 mg of 3-(8-carbamoyl-5,6,7,8-tetrahydro-[1,8]naphthyridin-3-yl)-isonicotinic acid.
