Procedure

To a solution of methyl 1-(2-oxo-2-((4-oxo-4,5,7,8-tetrahydro-3H-pyrano[4,3-d]pyrimidin-2-yl)methylamino)ethyl)piperidine-4-carboxylate (1.2 g, 3.3 mmol) in methanol (5 mL) was added sodium hydroxide (0.4 g, 10 mmol) in water (5 mL). The mixture was stirred at ambient temperature for 24 h, neutralized with HCl (37%) until pH 5˜7 and the solvent was evaporated under reduced pressure to give a white solid which was used directly to the next step. 1H NMR (400 MHz, CD3OD) δ 4.48 (s, 2H), 4.38 (s, 2H), 3.95 (t, J=5.8 Hz, 2H), 3.24-3.36 (m, 1H), 3.10 (d, J=12.1 Hz, 2H), 2.67 (t, J=5.6 Hz, 2H), 2.45 (td, J=11.4, 2.5 Hz, 2H), 2.25-2.39 (m, 1H), 1.77-2.04 (m, 5H).
