Procedure
To a stirred mixture of N-mustard amine 18 (7.66 g, 25 mmol) and triethylamine (TEA, (4 mL, 27.5 mmol) in CHCl3 (100 mL) was added a solution of 3-nitrophenyl isocyanate 44a (4.1 g, 25 mmol) in CHCl3 (50 mL) dropwise at room temperature. After being stirred additional 40-45 minutes, the solvent was removed by evaporation under reduced pressure and the solid residue was triturated with saturated aqueous solution of NaHCO3 and then collected by filtration. The solid was washed with water, hexane, and dried. The solid was recrystallized from ethyl acetate to give 1-(4-(bis(2-chloroethyl)amino)phenyl)-3-(3-nitrophenyl)urea (45a), 9.1 g (92%); mp 246-247° C. 1H NMR (DMSO-d6) δ: 3.69-3.71 (8H, m, CH2), 6.71-6.74 (2H, d, J=9.0 Hz, Ar—H), 7.29-7.31 (2H, d, J=8.9 Hz, Ar—H), 7.52-7.56 (1H, t, J=8.1 and 8.2 Hz, Ar—H), 7.69-7.71 (1H, d, J=8.0 Hz, Ar—H), 7.78-7.80 (1H, q, J=8.2 and 1.8 Hz, Ar—H), 8.52 (1H, brs, NH), 8.54-8.55 (1H, d, J=2 Hz, Ar—H), 9.11 (1H, brs, NH).
