Cells were harvested in 100 μl of 0,25 M Tris-HCl (pH 8); 80 μl were assayed for CAT activity by liquid scintillation counting using the same procedure as in in vitro translation assays, as described in the Technical Bulletin #084 from Promega, whereas 20 μl was used for assaying the β-galactosidase activity, with a colorimetric assay in which o-nitrophenyl β-D-galacto-side (ONPG) is cleaved to produce o-nitrophenol, as described in a standard protocol (Miller J. H. (1972) Experiments in Molecular Genetics, Cold Spring Harbor Laboratory Press, New-York, pp 352-355).
