0.85 g (5 mmol) of 2-aminoacetophenone hydrochloride and 1.98 g (25 mmol) of pyridine were dissolved in 30 ml of N,N-dimethylformamide in a four-necked flask equipped with a dry ice condenser. The solution was then cooled to a temperature of -50° C. To the solution was then added about 0.5 g (9.6 mmol) of cyanogen which had been cooled and solidified at a temperature of -50° C. The solution was heated at a temperature of 60° C. for 1 hour, and at a temperature of 80° C. for 1 hour. The reaction solution was then extracted with ethyl acetate and water. The resulting organic phase was then washed with saturated brine. The organic phase was dried over anhydrous sodium sulfate, and then concentrated. The material was then subjected to silica gel column chromatography to obtain 0.29 g (yield: 34 %) of 2-cyano-4-phenylimidazole.
