Procedure
1.5 ml of a 5M solution of L-threonine methyl ester, acetate containing 200 mg of porcine insulin were prepared by dissolving 200 mg of porcine insulin in 750 μ liters of 10M acetic acid and adding 1 g of L-threonine methyl ester thereto. 2 mg of porcine trypsin were added, and the clear reaction mixture having a pH value of 6.3 was left at 20° C. The reaction was stopped after 90 hours by adding 15 ml of water and adjusting the pH value to 3 with 5N hydrochloric acid.
