To N-(3-chloro-4-(difluoromethoxy)phenyl)-5-(2-(1-(tetrahydro-2H-pyran-2-yl)-1H-indazol-5-yl)ethyl)pyrimidin-2-amine (0.075 g, 0.150 mmol) in DCM (5 mL) was added TFA (0.116 mL, 1.500 mmol) and the reaction mixture was stirred at room temperature for 12 h. The reaction mixture was concentrated and the crude product purified by preparative HPLC on a Symmetry C18 column (19×300 mm, 7 μm) using 0-100% mobile phase B (ACN) in mobile phase A (10 mM ammonium acetate) to give 5-(2-(1H-indazol-5-yl)ethyl)-N-(3-chloro-4-(difluoromethoxy)phenyl)pyrimidin-2-amine as an off white solid (0.012 g, 19% yield). 1HNMR 400 MHz, DMSO-d6: δ 2.85 (t, J=15.60 Hz, 2H), 2.99 (t, J=14.80 Hz, 2H), 6.95-7.31 (m, 3H), 7.46 (d, J=8.40 Hz, 1H), 7.54 (s, 1H), 7.65-7.68 (m, 1H), 7.98 (s, 1H), 8.08 (d, J=2.80 Hz, 1H), 8.36 (s, 2H), 9.76 (s, 1H), 12.95 (s, 1H). LCMS (ES-API), RT: 2.15 min, m/z 414 (M−H).
