Trifluoroacetic acid (0.8 mL) was added dropwise to the cold solution of tert-butyl 4-((5-cyanopyridin-2-yl)methyl)-2-methylpiperazine-1-carboxylate (200 mg, 0.63 mmol) in CH2Cl2 (3 mL) at 0° C. and the reaction mixture was further stirred at room temperature for 2 h. The reaction mixture was concentrated under reduced pressure to afford 6-((3-methylpiperazin-1-yl)methyl)nicotinonitrile TFA salt (180 mg, crude), which was carried through without further purification. MS (ESI) m/z: Calculated for C12H16N4: 216.14. found: 217.0 (M+H)+
