Procedure
8-bromo-6-chloro-4-(piperidin-4-ylamino)-chromen-2-one and piperonal were allowed to react under reductive alkylation conditions for 44 hours at room temperature, then treated with 1 M K2CO3, extracted with CH2Cl2, filtered through silica gel (5 g SepPak), rinsed with 95:5 CH2Cl2:MeOH, and concentrated. Purification by silica gel preparatory plate (0.25 mm×20 cm×20 cm) developed with 95:5 CH2Cl2:MeOH to give the title compound as a white solid (6 mg). 1H NMR (300 MHz, ppm, DMSO-d6) δ 1.6 (m, 2H), 1.9 (m, 2H), 2.1 (m, 2H), 2.8 (m, 2H), 3.4-3.5 (m, 3H), 5.33 (s, 1H), 5.99 (s, 2H), 6.77 (m, 1H), 6.85 (m, 2H), 7.38 (m, 1H), 8.05 (d, J=2 Hz, 1H), 8.36 (d, J=2 Hz, 1H). MS(ESI) 493 (M+H), 491 (M−H).
