Dess-Martin Periodinane (50 mg, 0.12 mmol) was added to a suspension of 1-[4-[3-hydroxy-8-azabicyclo[3.2.1]octan-8-yl]-5-phenyl-thieno[2,3-d]pyrimidin-2-yl]pyrrolidine-3-carboxamide (40 mg, 0.089 mmol) in dichloromethane (4 mL). The mixture was stirred overnight at room temperature. Saturated aqueous sodium thiosulfate solution (2 mL) was added and the layers were separated. The aqueous was extracted with dichloromethane (2×2 mL). The combined organic layers were washed with saturated aqueous sodium bicarbonate solution (1×2 mL), filtered through a hydrophobic frit and evaporated to give the target compound as a white solid (46 mg). m/z [M+H]+448.1. Retention time 3.59 min (LCMS method+ve 6 min).
