A mixture of tert-Butyl-2-[(3,4,5-trimethoxyphenyl)amino]-5,6,7,8-tetrahydropyrido[4,3-d]pyrimidine-6-carboxylate, Intermediate 2 (870 mg, 2.09 mmol) in 4.0 N HCl solution in 1,4-dioxane (15 mL) was stirred at room temperature for 20 hours (orange colored solution with precipitate). The solvent was removed in vacuo and the salt was mostly taken up in water (20 mL). The pH of the aqueous mixture was made basic by addition of 1.0 N NaOH(aq) and the white, amorphous precipitate was collected, washed with water, ethyl acetate, and dried (616 mg, 93%).
