실험 절차

In a 50 mL round-bottom flask equipped with a stirring bar were placed PPh3 (0.823 g, 3.138 mmol) and I2 (0.800 g, 3.152 mmol) in dry CH2Cl2 (11.0 mL) and the mixture was stirred at room temperature for 20 min. DIPEA (1.5 mL, 8.62 mmol) and 3′,5′-bis-O-(tert-butyldimethylsilyl)-2′-deoxyinosine (1a) (0.500 g, 1.040 mmol) were added, and the mixture was allowed to stir at room temperature for 26 h. To this mixture was added HOBT (0.211 g, 1.561 mmol) and the reaction was allowed to continue and was complete in 22 h. The crude material was evaporated and chromatographic purification (SiO2, elution with 20% EtOAc in hexanes) afforded 0.578 g (93%) of compound 26 as a beige foam.
