To a pressure flask is added 6-amino-2,3-difluorobenzamide (0.700 g, 4.07 mmol), 2,4-dichloro-7-[(4-methylphenyl)sulfonyl]-7H-pyrrolo[2,3-d]pyrimidine (1.4 g, 4.07 mmol), trifluoroethanol (30 mL) and trifluoracetic acid (1.6 mL, 20.4 mmol). The resulting clear solution was stirred overnight. The next morning all precipitates were collected to afford analytically pure 6-({2-chloro-7-[(4-methylphenyl)sulfonyl]-7H-pyrrolo[2,3-d]pyrimidin-4-yl}amino)-2,3-difluorobenzamide as a white solid (1.07 g, 55% yield). 1H NMR (400 MHz, DMSO-d6) δ ppm 2.36 (s, 3 H), 6.74 (s, 1 H), 7.41-7.48 (m, 3 H), 7.49-7.60 (m, 1 H), 7.64 (d, J=3.67 Hz, 1 H), 7.82 (s, 1 H), 7.96 (d, J=8.43 Hz, 3 H), 10.13 (s, 1 H).
