A mixture of 1-(2-(3-methyl-1H-indazol-6-ylamino)furo[3,2-d]pyrimidin-4-yl)piperidine-4-carbonitrile (0.082 g, 0.22 mmol), hydroxylamine hydrochloride (0.061 g, 0.88 mmol) and NaHCO3 (0.15 g, 1.7 mmol) in MeOH (20 mL) was stirred at about 70° C. for about 120 h. The solvent was removed and the residue was purified by preparative HPLC (Table 2, Method p) to give N′-hydroxy-1-(2-(3-methyl-1H-indazol-6-ylamino)furo[3,2-d]pyrimidin-4-yl)piperidine-4-carboximidamide (0.020 g, 22%): LC/MS (Table 2, Method h) Rt=1.66 min; MS m/z: 407 (M+H)+.
