Into a round bottom flask containing N,N-diisopropylamine (2.692 mL, 19.21 mmol) in 50.0 mL or tetrahydrofuran, n-butyllithium (2.50 M in hexane, 7.684 mL, 19.21 mmol) was added at −78° C. under an atmosphere of nitrogen. After stirring at −78° C. for 30 minutes, N-(2,4-difluoro-phenyl)-4-trifluoromethyl-benzenesulfonamide (3, 2.09 g, 6.20 mmol) was added, with continued stirring at −78° C. under nitrogen for one hour. The reaction was allowed to come to room temperature over 15 minutes, then poured into water, acidified with 1 N aqueous hydrochloric acid to pH=1, and extracted with ethyl acetate. The organic layers were combined and washed with brine, then dried over sodium sulfate, filtered and the filtrate concentrated under vacuum. The resulting material was purified by silica gel column chromatography, the appropriate fractions were combined, and the solvents removed to provide the desired compound (4, 1.02 g). MS (ESI) [M−H+]−=364.1.
