A mixture of ethyl 5-(3-butyl-8-chloro-2,6-dioxo-2,3,6,7-tetrahydro-1H-purin-1-yl)pentanoate (2.8 g, 7.55 mmol), LiOH (542 mg, 22.7 mmol), water (2.5 ml) and methanol (50 ml) was stirred at rt for 60 h. The mixture was portioned between water and EtOAc and the pH of the aqueous phase adjusted to pH 4-5. The organic layer was separated, washed with brine, dried (MgSO4) and evaporated to give the title compound as a white solid (2.18 g).
