Procedure

The peptide is eliminated from the resin by treatment with about 10 ml of a (90:5:5) mixture of trifluoroacetic acid, water and 1,2-ethanedithiol for 8 minutes; the reaction mixture is filtered, the filtrate is concentrated under reduced pressure, and the peptide is precipitated with a 1:1 mixture of diisopropyl ether and hexane and dried. The abovementioned treatment with trifluoroacetic acid/water/1,2-ethanedithiol itself results in partial elimination also of the trityl groups (where present) and the tert.-butyl group. This elimination is completed by heating the peptide precipitate in trifluoroacetic acid/water/1,2-ethanedithiol (90:5:5) at 30° C., for 30 minutes. The resulting crude products from the syntheses with and without trityl protective groups are investigated by HPLC (column: Nucleosil 5C18, 250×4.6 mm; mobile phase: A=0.1% trifluoroacetic acid in water; B=0.1% trifluoroacetic acid in acetonitrile. Gradient: 0 to 40% B in 30 minutes; flow rate: 1 ml/minute. Detection: 215 nm).
