To a stirred solution of triphosgene (39 mg, 0.13 mmole) in dichloromethane (10 ml) was added a solution of 4-(pyridin-4-yl)naphth-1-ylamine (D1, 82 mg, 0.37 mmole) and triethylamine (0.05 ml, 0.37 mmole) dropwise over 30 minutes. When the addition was complete the mixture was stirred at room temperature for 15 minutes, then a solution of 4-chloro-3-(4-methylpiperazin-1-yl)aniline (100 mg, 0.44 mmole, EP0533268A1) in dichloromethane (10 ml) was added over 5 minutes. After 18 h, the mixture was washed with 10% aqueous sodium carbonate solution and water, dried (Na2SO4) and evaporated to dryness. The residue was purified by column chromatography on silica gel eluting with 10% methanol/dichloromethane and the title compound was obtained as a white solid on trituration with ether (47 mg, 27%).
