To a solution of 4-[4-(tert-butoxycarbonyl)piperazin-1-yl]-7-nitroquinoline (360 mg, 1 mmol) in CH2Cl2 (1 mL) is added trifluoroacetic acid (1 mL). After 1½ h at room temp., the mixture is concentrated, and the residue is dissolved in CH2Cl2 (20 mL) and treated with triethylamine (0.56 mL, 4 mmol) and 4-fluorophenyl isocyanate (0.16 mL, 1.4 mmol). After 1½ h stirring at room temp., the reaction mixture is washed with water and brine, dried (Na2SO4), and concentrated. The residue is purified by reversed phase HPLC with water-MeCN-TFA yielding the title product.
