Procedure
Acetic acid 2-acetoxymethyl-2-acetylamino-4-[4′-(5-propyl-isoxazol-3-yl)-biphenyl-4-yl]butyl ester (0.1 mmol) is dissolved in THF (1 mL) and treated with 2 N LiOH aqueous solution (0.5 mL). The resulting mixture is stirred at reflux for 1 h and diluted with H2O (10 mL). It is then extracted with EtOAc (3×5 mL) and the combined organic phase is washed with brine and dried over Na2SO4. After concentrated, the crude product is purified with LC-MS to give the desired product as a white solid. 1H NMR (400 MHz, CD3OD) δ 7.86 (d, J=8.4 Hz, 2H), 7.70 (d, J=8.4 Hz, 2H), 7.58 (d, J=8.2 Hz, 2H), 7.33 (d, J=8.2 Hz, 2H), 3.53 (q, J=11.0 Hz, 4H), 2.81 (t, J=7.4 Hz, 2H), 2.71 (m, 2H), 1.79 (m, 4H), 1.04 (t, J=7.4 Hz, 3H). MS: (ES+): 381.2 (M+1)+.
