A suspension of crude 5-chloro-1-isobutyl-3-methyl-1H-pyrrolo[3,4-d]pyrimidine-2,4(3H,6H)-dione (0.032 mmol), 2-(4-(chloromethyl)phenyl)pyridine (9.7 mg, 0.048 mmol) and cesium carbonate (15.6 mg, 0.048 mmol) in anhydrous DMF is stirred at room temperature for 3 h, and then diluted with saturated NaHCO3 aqueous solution, followed by extractions with CH2Cl2. The combined organic phase is dried over sodium sulfate, filtered, and concentrated to dryness under reduced pressure to give crude product, which is used in the next step without further purification. MS (ESI) m/z 423.1 [M+H]+.
