Procedure
To a solution of 5-(4-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-ethynylpyrimidin-2-yl)thiophene-2-carbonitrile (80 mg, 0.24 mmol, 1.0 equiv.) in MeOH (5 mL) was added K2CO3 (133 mg, 0.96 mmol, 4.0 eq). After being stirred at 20° C. for 4 h, HCl (10%, 5 mL) was added, and the reaction mixture was stirred at 20° C. for 30 min. The reaction mixture was extracted with ethyl acetate, the organic layer was washed with water, dried (Na2SO4) and evaporated and crystallized with petroleum ether/ethyl acetate to afford the product compound methyl 5-(4-(5-cyclopropyl-1H-pyrazol-3-ylamino)-5-ethynylpyrimidin-2-yl)thiophene-2-carboxylate hydrochloride (Compound 151 (21.2 mg, 22%). LC-MS (m/z)=366 [M+H]+. 1H NMR (400 MHz, DMSO-d6): δ 0.75-0.79 (m, 2H), 0.99-1.02 (m, 2H), 1.95-1.99 (m, 1H), 3.87 (s, 3H), 4.92 (s, 1H), 6.46 (s, 1H), 7.85 (dd, J=4.0 Hz, J=3.6 Hz, 2H), 8.54 (s, 1H), 8.99 (s, 1H).
