To a solution of (S)-1-chloro-4-(3-methylpiperazin-1-yl)pyrido[3,4-d]pyridazine 4 (500 mg, 1.90 mmol) in 10 mL of CH2Cl2 was added triethylamine (529 μl, 3.79 mmol), followed by 1-piperidinecarbonyl chloride (364 μl, 2.47 mmol). The reaction was allowed to stir overnight and then diluted with 100 mL of ethyl acetate. The crude mixture was washed with 1×10 mL of sat. NaHCO3, 1×10 mL of water, and 1×10 mL of brine. The organic layer was dried over MgSO4, filtered, and concentrated. Purification by column chromatography afforded (S)-(4-(1-chloropyrido[3,4-d]pyridazin-4-yl)-2-methylpiperazin-1-yl)(piperidin-1-yl)methanone 7.
