A suspension of 4-(3-hydroxy-9-phenyl-[1,2,4]triazolo[3,4-f][1,6]naphthyridin-8-yl)-benzaldehyde (3-6, 2.4 g, 6.6 mmol) and 2-amino-2,4-dimethylpentanoic acid (3-7, 1.9 g, 13.1 mmol) in DMF (25 mL) was heated at 150° C. for 20 min. The reaction mixture was cooled, diluted with dioxane (50 mL) and 2N HCl (50 mL) and heated at 100° C. for 30 min. The mixture was cooled and purified via reverse phase HPLC (0-60% acetonitrile/water) to give, upon evaporation of the solvent, the title compound as a yellow solid. HRMS m/z (M+1) Calcd: 368.1506. Found 368.1504.
