Procedure
To a solution of ((1S,3′R,4′S,5′S,6′R)-5-chloro-6-(4-iso-propylbenzyl)-3′,4′,5′-trihydroxy-3′,4′,5′,6′-tetrahydro-3H-spiro[isobenzofuran-1,2′-pyran]-6′-yl)methyl 4-methylbenzenesulfonate (15 mg, 0.025 mmol) and dimethylamino hydrochloride (48 mg, 0.588 mmol) in DMF (1 mL), was added Et3N (67 mg, 0.662 mmol). After being stirred for 24 h at 80° C., the reaction mixture was diluted with H2O and extracted 3× with AcOEt. The combined organic layers were washed with brine prior to drying over Na2SO4. The volatiles were removed under reduced pressure. The resulting residue was purified by preparative HPLC to yield 6 mg of title compound. 1H NMR (CD3OD, 300 MHz) δ 7.41 (s, 1H), 7.24 (s, 1H), 7.13 (s, 4H), 5.18 (d, J=13.5 Hz, 1H), 5.13 (d, J=13.5 Hz, 1H), 4.17-4.02 (m, 3H), 3.76-3.75 (m, 2H), 3.46-3.18 (m, 3H), 2.90-2.81 (m, 1H), 2.76 (s, 6H), 1.22 (d, J=6.9 Hz, 6H); MS ESI (m/z) 462 (M+1)+, calc. 461.
