实验过程

A mixture of intermediate 21 (220 mg, 0.74 mmol) and NaI (222 mg, 1.48 mmol) in CH3CN was heated to reflux for 30 min and then cooled to rt. Intermediate 41 (278 mg, 1.04 mmol) and anhydrous K2CO3 (1409 mg, 2.96 mmol) were added to the mixture. The resulting mixture was heated to reflux and stirred for 4 h. Precipitated crystals were filtered off and the filtrate was evaporated under reduced pressure. The residue was extracted with EtOAc. The combined EtOAc layers were washed with brine, dried over anhydrous Na2SO4, concentrated in vacuo and purified by flash chromatography on silica gel column (elution with DCM/MeOH=50:1) to give 7-(4-(4-(2,3-dichlorophenyl)piperazin-1-yl)butoxy)-3,4-dihydroisoquinolin-1(2H)-one (compound 25) (215 mg, 65%). 1H NMR (300 MHz, CDCl3): δ 7.58 (d, J=2.7 Hz, 1H), 7.14-7.11 (m, 3H), 7.01-6.94 (m, 2H), 6.18 (brs, 1H), 4.05 (t, J=6.3 Hz, 2H), 3.56-3.51 (m, 2H), 3.07 (m, 4H), 2.90 (t, J=8.1 Hz, 2H), 2.66 (m, 4H), 2.49-2.46 (m, 2H), 1.86-1.68 (m, 4H). HPLC: 99%, RT 2.374 min. MS (ESI) m/z 448.3 [M+H]+.
