A mixture of tert-butyl 4-oxo-1-phenyl-1,3,8-triazaspiro[4.5]decane-8-carboxylate (2 g, 6.02 mmol, 1 equiv), methyl 3-(bromomethyl)benzoate (1.38 g, 6.02 mmol, 1 equiv) and potassium carbonate (1.25 g, 9.03 mmol, 1.5 equiv) in N,N-dimethylformamide was heated at 65° C. for 16 h. Upon cooling the mixture was partitioned between ethyl acetate and water. The organic layer was further washed with brine, dried over MgSO4, filtered and concentrated in vacuo. The residue was purified using the Biotage flash chromatography system (SNAP 50 g cartridge, Rf=0.5, gradient—10%-50% ethyl acetate in hexanes) to afford tert-butyl 3-(3-(methoxycarbonyl)benzyl)-4-oxo-1-phenyl-1,3,8-triazaspiro[4.5]decane-8-carboxylate as a white solid (1.67 g, 58%); MS for C27H33N3O5 m/z 479.24 (M+H)+.
