A mixture of 3-(1-(2,6-dichloro-3-fluoro-phenyl)ethoxy)-5-(1-piperidin-4-yl-1H-pyrazol-4-yl)-pyridin-2-amine (6 mg, 0.013 mmol), chloromethyl-dimethyl-phosphine oxide (100 mg, 0.79 mmol), and potassium carbonate (0.2 g) in DMF (5 mL) was heated to 80° C. overnight. The solid was removed and the residue was purified by HPLC (water/methanol, 10˜100%) to give the title compound as an off-white solid (3 mg, 40%); ESMS: m/z 540 (M+H)+.
