Procedure

A mixture of 1-(tert-butoxycarbonyl)-2-methylproline (1 eq), TEA (2 eq), PyBOP (1 eq) and 6-[4-fluoro-3-(piperazin-1-ylcarbonyl)benzyl]-4,5-dimethylpyridazin-3(2H)-one (II) in dry DMF (0.1 M) was heated at 60° C. with microwaves irradiation for 2 hours. The product was isolated by preparative-HPLC. After lyophilization of the pooled product fractions the residue was dissolved in 10% TFA/DCM and the solution was warmed to 45° C. for 1 h. The residue obtained after solvent evaporation was further purified by SCX cartridge. Lyophilization from water/MeCN afforded the title compound as a white powder. 1H NMR (400 MHz, DMSO-d6) δ: 12.67 (1H, s), 7.38-7.13 (3H, m), 3.96 (2H, s), 3.8-3.5 (5H, m), 3.3 (1H under water signal), 3.26-3.13 (2H, m), 2.92-2.78 (1H, m), 2.76-2.58 (1H, m), 2.34-2.15 (1H, m), 2.02 (3H, s), 2.00 (3H, s), 1.75-1.46 (3H, m), 1.26 (3H, s). MS (ES) C24H30FN5O3 requires: 455. found: 456 (M+H)+.
