Procedure
A 20-mL vial was charged with N-(((2R)-4-((5-nitro-2-thiophenyl)sulfonyl)-1-(5-(2,2,2-trifluoro-1-hydroxy-1-(trifluoromethyl)ethyl)-2-pyrimidinyl)-2-piperazinyl)methyl)-N-phenylmethanesulfonamide (0.131 g, 0.186 mmol, step 1), iron filings (0.052 g, 0.93 mmol) and acetic acid (2 mL). The reaction mixture was heated at 50° C. for 40 min and then allowed to cool to room temperature. Saturated aqueous NaHCO3 (20 mL) was then added slowly. The aqueous phase was extracted with EtOAc (2×20 mL) and the combined organic phases were washed with saturated aqueous NaCl (30 mL), dried over sodium sulfate, filtered and concentrated in vacuo. The crude product was purified by column chromatography (25 g of silica gel, 20 to 80% EtOAc in hexanes) to afford N-(((2R)-4-((5-amino-2-thiophenyl)sulfonyl)-1-(5-(2,2,2-trifluoro-1-hydroxy-1-(trifluoromethyl)ethyl)-2-pyrimidinyl)-2-piperazinyl)methyl)-N-phenylmethanesulfonamide (0.0912 g) as a light-yellow solid.
