Procedure
Methyl 4-(9-cyclopentyl-7,7-dimethyl-6-oxo-6,7,8,9-tetrahydro-5H-pyrimido[4,5-b][1,4]diazepine-2-ylamino-3-methoxybenzoate and dicesium carbonate (118.6 mg, 0.3640 mmol) were mixed in DMF. MeI (103.3 mg, 45.31 μL, 0.7280 mmol) was then added and the reaction mixture was stirred at room temperature overnight. The mixture was diluted with ethylacetate, filtered through small pad of celite, washed twice with water, dried and concentrated to a yellow solid. Purification by flash chromatography on silica gel, eluting with 1% methanol/DCM, gave methyl 4-(9-cyclopentyl-5,7,7-trimethyl-6-oxo-6,7,8,9-tetrahydro-5H-pyrimido[4,5-b][1,4]diazepin-2-ylamino)-3-methoxybenzoate as a pale yellow solid (47 mg, 56%). NMR CDCl3 1.25 (6H, s), 1.53-1.58 (2H, m), 1.69-1.83 (4H, m), 2.02-2.08 (2H, m), 3.33 (3H, s), 3.4 (2H, s), 3.92 (3H, s), 3.98 (3H, s), 5.27-5.34 (1H, m), 7.23-7.26 (1H, brs), 7.55 (1H, s), 7.72-7.77 (2H, m), 7.88 (1H, s), 8.56 (1H, d); MS (ES+) 454.1 (ES−) 452.1
