2,4-Dichloro-7-iodo-quinoline (3.4 g, 10.5 mmol), 4-(3-Mercapto-phenyl)-tetrahydro-pyran-4-carbonitrile (5q, 2.3 g, 10.5 mmol), iPr2NEt (3.67 mL, 21.0 mmol), Pd2dba3 (240 mg, 0.26 mmol), and 4,5-bis(diphenylphosphino)-9,9-dimethylxanthene (303 mg, 0.52 mmol) were dissolved in 1,4-dioxane (50 mL) and degassed with N2 for 10 minutes. The reaction was heated at 60° C. for 1 hour, and then cooled to room temperature and diluted with EtOAc and saturated aqueous NH4Cl. The aqueous layer was extracted with EtOAc, and the combined organic layers were dried over MgSO4, filtered, and concentrated. The residue was purified by silica gel chromatography (30% EtOAc in hexanes) to give the desired product, 5r.
