Procedure
PAMAM generation 2.0 core (0.0479 mmol; 50 mg) was evaporated from a 0.5 ml solution in MeOH and then re-dissolved in 10 ml of water. 1-N-pyridinium 12-dodecanoic acid bromide (0.14 g; 0.384 mmol), N-hydroxybenzotriazole hydrate [HOBT] (52 mg; 0.384 mmol); triethylamine (531 0.384 mmol) and 1-(3-diethylaminopropyl-3-ethyl) carbodiimide .HCl [EDC] (74 mg; 0.384 mmol), were added to the solution. This reaction mixture was stirred overnight at room temperature. The volume was reduced to a third under reduced pressure and the solution was chromatographed on a LH20 column using water as the eluent. Fractions containing the product were collected and pyridinium dodecylcarboxamide PAMAM 2.0 bromide isolated as a fluffy white solid by freeze drying.
