Procedure

Triethylamine (0.26 ml), 2-chlorobenzyl chloroformate ([39545-31-8], 130 mg) and DMAP (8 mg) were added to a solution of 2-aminomethyl-3H-pyrido[2,3-d]pyrimidin-4-one (25.1, 150 mg) in dichloromethane (3 ml). The mixture was kept overnight at r.t., and then diluted with dichloromethane and washed with water. After drying (Na2SO4) and evaporation of the solvent, the title compound was isolated from the reaction mixture by preparative, reversed-phase HPLC (Agilent Zorbax XdB C18 column, solvent gradient 5-95% CH3CN in 0.1% TFA(aq) over 7 min, flow rate 30 ml/min). MS: m/e=343.0 [M−H−].
