To a solution of Intermediate 415.2 (115 mg, 0.171 mmol) in 2 mL THF at rt, was added TBAF (1 M in THF, 171 μL, 0.171 mmol). The mixture was stirred at rt for 20 min, then was diluted with EtOAc, washed with H2O (2×) and brine, dried (Na2SO4) and concentrated. The crude residue was purified by flash chromatography (0 to 80% EtOAc/hexanes gradient) to afford 34 mg of 3-((5,6-dimethyl-1-trityl-1H-benzo[d]imidazol-2-yl)(hydroxy)methyl)-5-ethyl-2-fluorophenol as a white solid.
