To a mixture of 2-[(2-chloro-5-nitropyrimidin-4-yl)amino]benzamide (52 mg, 0.178 mmol) and 3,4,5-trimethoxyaniline (34 mg, 0.185 mmol) in 2-propanol (2 mL) was added 1N HCl in diethyl ether (50 uL, 0.05 mmol). The reaction mixture was heated at 90° C. for approx. 18 hours. After the reaction was cooled to room temperature cold diethyl ether was added to precipitate out the product. The precipitate was filtered and washed with cold diethyl ether, then dried under vacuum to give 2-({5-nitro-2-[(3,4,5-trimethoxyphenyl)amino]pyrimidin-4-yl}amino)benzamide hydrochloride (30 mg, 35%) as a dark brown solid. 1H NMR (DMSO-d6/D2O): δ 9.07 (s, 1H), 8.26 (m, 1 H), 7.69 (d, 1 H), 7.34 (t, 1 H), 7.19 (t, 1 H), 6.92 (s, 2 H), 3.77 (s, 3 H), 3.59 (s, 6 H). MS m/z 440 (M+1).
