Lithium hydroxide (965 mg, 23.0 mmol) was added to a suspension of methyl 4-(5-(3-cyanophenyl)-1,3-dimethyl-2,4-dioxo-3,4-dihydro-1H-pyrrolo[3,4-d]pyrimidin-6(2H)-yl)butanoate (Step 5) (1.75 g, 4.6 mmol) in THF (9 mL)/water (9 mL). The mixture was stirred at room temperature for 4 hours. The organic solvent was largely removed under vacuum, the residue was acidified to pH 1, using 2M HCl. The resulting slurry was diluted with water (100 mL). The mixture was reduced under vacuum to give a white solid, which was further dried under vacuum at 50° C. overnight.
