To a mixture of 6-Bromo-4-{[4-(2-hydroxymethyl-pyrrolidin-1-ylmethyl)-phenylamino]-methylene}-4H-isoquinoline-1,3-dione (100 mg, 0.2 mmol) in N,N-dimethylformamide (1 mL) is added Pd2(dba)3.CHCl3 (10 mg, 0.01 mmol) and P(tBu)3 (20 μL, 1 M solution in N,N-dimethylformamide, 0.02 mmol). After degassing the mixture, 2-thienyl zinc chloride (2.2 mL, 0.5 M solution in THF, 1.1 mmol) is then added and the resulting mixture is stirred for 2 h. N,N-dimethylformamide is then removed and the residue is purified through chromatography to provide the title compound (35 mg, 38%). MS (ESI): 460.1 (M+1)+1.
