PROCEDURE
实验过程
A solution of 61 mg (0.88 mmol) of hydroxylamine hydrochloride in 1 ml of H2O is mixed with a solution of 56 mg (1.4 mmol) of NaOH in 0.5 ml of H2O and stirred for 10 minutes. At 0° C., a solution of 240 mg (0.4 mmol) of methyl 2-[4-(4-fluorophenoxy)-benzenesulfonyl]-6-[3-(4-methylpiperazin-1-yl)propoxy]-1,2,3,4-tetrahydroisoquinoline-1-carboxylate in 0.5 ml of dioxane is added, and the mixture is stirred at 0° C. To achieve complete conversion, 61 mg (0.88 mmol) of hydroxylamine hydrochloride and 56 mg (1.4 mmol) of NaOH are added twice more. For work up, the mixture is neutralized with 1 N HCl and extracted with ethyl acetate. The organic phase is separated off, dried with MgSO4 and concentrated. Chromatography of the residue on silica gel affords the hydroxamic acid (exemplary compound 3) as colorless solid in 46% yield, and the carboxylic acid (exemplary compound 4), likewise as colorless solid in a yield of 15%.
