N-[5-chloro-7-(trifluoromethyl)[1,2,4]triazolo[1,5-a]pyridin-2-yl]nicotinamide ((B7), 50 mg; 0.14 mmol; 1.0 eq.) in cyclopentylamine (1.0 mL) was stirred at rt overnight. The reaction mixture was diluted with a saturated solution of NaHCO3 (10 mL) and EtOAc (5 mL). The two phases were separated and the organic phase was washed four times with a saturated solution of NaHCO3, after which the product precipitated in the organic phase. It was filtered, washed with a saturated solution of NaHCO3 and EtOAc, dried under vacuum and isolated as a white solid (13 mg, 23%). HPLC, Rt: 3.54 min. (purity 100%). LC/MS, M+(ESI): 391.3, M−(ESI): 389.3.
