Procedure
To a stirred solution of 250 mg of methyl 1,4-dihydro-2,6-dimetyl-3-[3-(N-benzyl-N-methylamino)methyl-1,2,4-oxadiazol-5-yl]-4-(3-nitrophenyl)pyridine-5-carboxylate in 5 ml of dry DMF was added 24 mg of 60% sodium hydride with ice-cooling. After the mixture was stirred for 10 minutes, there was added 76 mg of methyl iodide. The reaction mixture was allowed to stand at room temperature for 6 hours, diluted by addition of ice-water and then extracted with ethyl acetate. The extracts were washed with water, dried and evaporated in vacuo. The residue was chromatographed on silica gel using chloroform-methanol (30:1) as eluent to give 190 mg of methyl 1,4-dihydro-3-[3-(N-benzyl-N-methylamino)methyl-1,2,4-oxadiazol-5-yl]-1,2,6-trimethyl-4-(3-nitrophenyl)pyridine-5-carboxylate, IR νmaxneat cm-1 : 1690, 1635, 1580, 1520, 1340, 900.
