Potassium fluoride (72.5 mg, 1.25 mmol) was added to a stirred solution of 5-(2-butoxy-5-trimethylsilylethynyl-3-pyridinyl)-2-[2-(dimethylamino)-ethyl]-3-ethyl-2,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-one (Example 45) (300 mg, 0.625 mmol) in N,N-dimethylformamide (10 mL) and water (2 mL) at room temperature. After 2 h the reaction mixture was poured into brine and extracted with ethyl acetate (2×100 mL) The organics were dried (MgSO4) and concentrated to give the product (285 mg) as a pale brown oil.
