Procedure

A solution of N-(4-hydroxyphenyl)-3-pyridylmethylamine (1.0 equiv.) in DMF (0.1-0.3M) was treated with sodium hydride (1.0-1.5 equiv.)(60% dispersion in mineral oil). The reaction mixture was allowed to stir at room temperature for 30 minutes. The solution was then treated with 3,4-difluorobenzonitrile (1.6 equiv.) and allowed to stir. When complete (about 3 hours) the reaction mixture was partitioned between ethyl acetate/0.1M K2CO3, the organic layer separated, and the aqueous layer extracted 3 times with ethyl acetate. The organic layers were combined, washed with brine, and dried over MgSO4. The organic phase was concentrated in vacuo and purified by recrystallization from ethyl acetate/hexanes to give N-(4-(2-fluoro-4-cyanophenoxy)phenyl)-3-pyridylmethyl-amine 1H NMR(400 MHz, DMSO-d6): 4.29(2H, d, J=5.9 Hz), 6.40(1H, t, J=5.5 Hz), 6.63-6.65(2H, m), 6.83(1H, t, J=8.6 Hz), 6.90-6.92(2H, m), 7.33-7.36(1H, m), 7.55-7.57(1H, m), 7.74-7.76(1H, m), 7.93-7.97(1H, m), 8.43-8.44(1H, m), 8.58(1H, s). MS Found 320.1 [M+H]+.
