1,1′-Carbonyldiimidazole (25 g, 155 mmol) was added to a solution of 3-[(3-amino-2-chloroquinolin-4-yl)amino]propan-1-ol (26 g, 0.010 mol) in THF (140 mL), and the reaction was stirred for three hours at room temperature. Additional 1,1′-carbonyldiimidazole (25 g) and pyridine (50 mL) were added, and the reaction was heated at 90° C. for three hours and allowed to cool. The reaction was cooled to 0° C., and water (300 mL) was added. The resulting mixture was stirred for 30 minutes, and a precipitate formed. The precipitate was isolated by filtration and washed with water to provide 38 g of 3-(4-chloro-2-hydroxy-1H-imidazo[4,5-c]quinolin-1-yl)propyl 1H-imidazole-1-carboxylate as a white solid.
