Procedure
To a mixture of 2-amino-5-bromo-4-chloro-3-nitropyridine (0.126 g, 0.50 mmol) and isopropanol (6 ml) was added 1-cyclopropylmethylpiperazine (0.078 g, 0.55 mmol) with the aid of isopropanol (3 ml), followed by diisopropylethylamine (0.10 ml, 0.57 mmol). The reaction mixture was stirred at 45° C. for 20 h, then allowed to cool to room temperature and diluted with isopropanol (4 ml). The resulting yellow precipitate was collected by filtration, washed with isopropanol (2×4 ml), diethyl ether (3×4 ml), and dried. The title compound was obtained as a yellow solid (0.096 g). The filtrate was concentrated in vacuo, and purification of the resulting residue by chromatography (10 g isolute column) eluting with a gradient of methanol (2 to 4%) in dichloromethane/ethyl acetate (v/v; 1:1) afforded an additional 0.038 g of the product (overall yield: 76%). 1H-NMR (500 MHz, DMSO-d6) 0.08 (m, 2H) and 0.46 (m, 2H), (cyclopropyl CH2), 0.86 (m, 1H, cyclopropyl CH), 2.22 (d, J=6.6 Hz, 2H, N—CH2cyclopropyl), 2.57 (br s, 4H, piperazine N(CH2)2), 3.06 (br s, 4H, piperazine N(CH2)2), 6.96 (s, 2H, NH2), 8.15 (s, 1H, pyridine 6-H);
