Phosphate → D-(-)-Fructose
Reaction equation
[O:1]=[CH:2][C@@H:3]([C@H:5]([C@@H:7]([C@@H:9]([CH2:11][OH:12])[OH:10])[OH:8])[OH:6])[OH:4].P([O-])([O-])([O-])=O>>[OH:1][CH2:2][C:3]([C@H:5]([C@@H:7]([C@@H:9]([CH2:11][OH:12])[OH:10])[OH:8])[OH:6])=[O:4]Participants

Procedure
To a glucose isomerase solution are added 0.1 M D-glucose, 0.05 M phosphate buffer solution and 0.005 M MgSO4.7H2O and, while maintaining pH at 7.0, the reaction is carried out at 70° C. for one hour. The amount of fructose formed is measured. One unit of activity is expressed as the amount of enzymes necessary for forming 1 mg of fructose under the above conditions.
