A mixture of 6-amino-5-chloro-2-(methyloxy)-N-(4-piperidinylmethyl)-3-pyridinecarboxamide (step 1, 150 mg, 0.50 mmol), (bromomethyl)cyclohexane (107 mg, 0.60 mmol), and triethylamine (140 μl, 1.00 mmol) in N,N-dimethylformamide (5 ml) was stirred at 90° C. for 10 h. After cooling to room temperature, the mixture was poured into water (50 ml) and the aqueous layer was extracted with ethyl acetate/toluene (2:1, 80 ml×2). The combined organic layer was washed with brine (100 ml), dried over magnesium sulfate and concentrated in vacuo. The residue was chromatographed on a column of silica gel eluting with 25% ammonia/methanol/dichloromethane (0.1:1:20) to give 130 mg (65%) of the title compound as a yellow amorphous solid.
