Procedure
A mixture of 2-methyl-5-(4-morpholinyl)-3-(1-naphthalenylmethyl)pyrazolo[1,5-a]pyrimidine-7-carboxamide (55 mg, 0.137 mmol) in N,N-dimethylformamide dimethyl acetal (2 mL, 14.94 mmol) was stirred at 105° C. for 3 days and concentrated to give the crude. To the crude was added Acetic Acid (2 mL) and hydrazine hydrate (0.047 mL, 0.959 mmol). The reaction mixture was heated at 100° C. for 1 h and concentrated. Brine (10 mL) and EtOAc (20 mL) was added in. The aqueous phase was extracted with EtOAc (20 mL×4). The organic phases were combined and concentrated. The crude was purified on silica column (EtOAc/Hexane: 10˜40%) to give the product (20 mg, 33%). 1H NMR (400 MHz, DMSO-d6) δ ppm 2.27 (s, 3H) 3.56-3.82 (m, 8H) 4.39 (s, 2H) 7.14 (s, 1H) 7.29-7.59 (m, 4H) 7.77 (d, J=8.08 Hz, 1H) 7.85-7.98 (m, 1H) 8.38-8.61 (m, 2H); LC/MS: MS (ES+) m/e 426.0 [M+H]+.
