6-Methoxy-8-methyl-4-(4-oxo-1-phenyl-1,3,8-triazaspiro[4.5]dec-8-yl)pteridin-7(8H)-one (1 g, 2.37 mmol) was treated with lithium hydroxide monohydrate (0.281 g, 4.74 mmol) tetrahydrofuran (2.5 mL) and water (2.5 mL). This mixture was stirred at 60° C. for 48 hours. The reaction mixture was cool to room temperature acidified to pH 4 with 1 M hydrochloric acid (8 mL) and evaporated under reduced pressure. The resulting crude material was then used in the next step without any further purification. MS (EI) for C20H21N7O3: 408 (MH+), 364 (MH+−44 (—CO2)).
